在室温下,向搅拌的2,6-二氯吡啶(68.9 g,466 mmol)的甲醇(500 mL)溶液中缓慢加入甲醇钠(100 g,1.86 mol)。将反应混合物在60°C下持续搅拌24小时。反应完成后,将混合物冷却至室温,用2 M盐酸水溶液淬灭反应,并用二氯甲烷(CH2Cl2)进行萃取。合并有机层,用无水硫酸镁(MgSO4)干燥,过滤后减压浓缩,得到2-氯-6-甲氧基吡啶(66.9 g,定量收率),为无色油状物。产物表征数据如下:IR(薄膜法,cm?1):1599, 1585, 1560, 1468, 1410, 1302, 1265, 1152, 1024, 876, 789;1H-NMR(CDCl3, 500 MHz)δ:7.51(t,J = 7.37 Hz,1H),6.90(d,J = 7.37 Hz,1H),6.65(d,J = 7.37 Hz,1H),3.94(s,3H);13C-NMR(CDCl3, 125 MHz)δ:163.9, 148.4, 140.5, 116.2, 109.1, 54.0;HR-MS(ESI-TOF):C6H7ClNO [(M + H)+]的计算值为144.0211,实测值为144.0211。
