以5,6-二氯-3-嘧啶甲醇为原料合成2,3-二氯-5-甲酰基吡啶的一般步骤:向500mL圆底烧瓶中加入2,3-二氯-5-羟甲基吡啶(64.810g,50.0mmol)的无水二氯甲烷(150mL)溶液,随后加入氧化锰(43.5g,0.50mol)。将反应混合物在25℃下搅拌48小时。反应完成后,通过Celite过滤,滤液减压浓缩。所得粗产物通过硅胶柱色谱纯化,采用乙酸乙酯(0%-40%)/己烷梯度洗脱,得到2,3-二氯-5-甲酰基吡啶(7.2g,产率90%)。产物经1H NMR(400MHz,CDCl3)确认:δ10.08(1H,s),8.77(1H,d,J=1.97Hz),8.25(1H,d,J=1.97Hz)。LC/MS分析显示分子离子峰(M+1)为176。
