一般步骤:以2-(羟甲基)丙烯酸甲酯为原料合成(-)-甲基-D-BETA-羟基异丁酸酯的一般程序如下:Ene-还原酶活性测定需在环境温度下于厌氧条件(Coy Laboratory,Grass Lake,MI)中进行。反应原液的制备包括将各底物(具体见下文)溶解于50mM Tris-HCl缓冲液(pH7.5)中,并补充200μM NADP+、100mM葡萄糖及来自Thermoplasma acidophilum的葡萄糖脱氢酶(GDH)(IVTT反应使用2个单位,纯化酶反应使用5个单位)。选择单个底物浓度以确保达到vmax条件或最大溶解度。在这些底物浓度下,调整反应时间以达到10-50%的底物转化率。对于IVTT实验中的OYE活性测定,将20μL反应原液与2-10μL IVTT反应混合物混合,并用50mM Tris-HCl(pH7.5)将总测定体积调整至30μL。对于纯化的OYE1变体的测定,将酶(最终浓度:250nM)加入至500μL反应储备溶液中。通过从测定溶液中取出30μL等分试样,并与30μL含有1mM环己酮(作为内标)的乙酸乙酯充分混合以淬灭反应进程。收集有机相样品并通过气相色谱(GC)进行分析(具体方案见下文)。通过积分产物和底物的峰面积来计算对映体/非对映异构体过量。各底物的相对转化率通过将测得的OYE1变体的转化率除以野生型OYE1的相应速率来计算。
