以甲醇和5-溴-2-氯烟酸为原料合成5-溴-2-氯烟酸甲酯的一般步骤如下:将浓硫酸(2 mL,52.1 mmol)缓慢加入至5-溴-2-氯烟酸(12.3 g,52 mmol)在甲醇(100 mL)中的悬浮液中。将反应混合物在搅拌下加热回流15小时。反应完成后,通过旋转蒸发仪除去溶剂。将残余物用饱和碳酸氢钠水溶液中和至pH 7-8。过滤所得悬浮液,用水洗涤固体产物,并通过真空泵干燥。最终得到5-溴-2-氯烟酸甲酯(11.6 g,收率89%)。产物经核磁共振氢谱(400 MHz,CDCl3)确认:δ 8.53(d,J = 2.4 Hz,1H),8.25(d,J = 2.4 Hz,1H),3.93(s,3H)。核磁共振碳谱(101 MHz,CDCl3):δ 163.58, 152.75, 148.61, 142.49, 127.64, 118.57, 53.10。电喷雾质谱(ESI-MS)显示m/z:251.9([M+H]+)。
