The general procedure for the synthesis of 6-methyl-1-(phenylsulfonyl)-1H-pyrrolo[2,3-b]pyridine from 6-chloro-1-(phenylsulfonyl)-1H-pyrrolo[2,3-b]pyridine and zinc methyl chloride was as follows: a THF solution of zinc methyl chloride (2M, 9 mL, 12 mmol) was slowly added to a solution containing 6-chloro-1-(phenylsulfonyl)-1H-pyrrolo[2,3-b]pyridine (0.600 g, 2.05 mmol) and Pd(PPh3)4 (0.095 g, 0.05 mmol). [2,3-b]pyridine (0.600 g, 2.05 mmol) and Pd(PPh3)4 (0.095 g, 0.08 mmol) in a THF solution (30 mL). The reaction mixture was stirred under reflux conditions for 40 h. The reaction was subsequently cooled to 0 °C and quenched with deionized water. The reaction mixture was extracted with ether (Et2O) and the organic layers were combined and concentrated. The crude product was purified by silica gel column chromatography with ethyl acetate/hexane (1:5, v/v) as eluent to afford the target product 6-methyl-1-(phenylsulfonyl)-1H-pyrrolo[2,3-b]pyridine (0.495 g, 88% yield).
