Summary:

Exemestane is an irreversible steroidal aromatase inhibitor with a structure similar to its substrate natural androstenedione. Estrogen deprivation by inhibiting aromatase is an effective and alternative treatment for hormone-dependent breast cancer in postmenopausal women with advanced breast cancer that has not progressed after tamoxifen therapy in natural or artificial postmenopausal women.

Uses:

It is an irreversible steroidal aromatase inactivator, which can inhibit estrogen synthesis and reduce the level of estrogen in blood circulation. It is suitable for advanced postmenopausal breast cancer patients who progress after tamoxifen treatment

Production method: 

Androstene-4-ene-3, 17-dione and triethyl orthoformate are dissolved in tetrahydrofuran and anhydrous ethanol, react in the presence of p-toluenesulfonic acid, and then add n-methylbenzene Chemicalbook amine and formaldehyde solution to continue the reaction. The Mannich reaction product was dissolved in benzoic acid and anhydrous dichethanes, and exemestane was obtained under the action of DDQ with a total yield of 45%.

Synthesis: 6-methylene-androstene-4-ene-3, 17-diketone (3) was synthesized from androstene-4-ene-3, 17-diketone (3) by condensation with triethyl orthoformate, N-methylaniline and 40% formaldehyde solution, and then 1 was prepared by oxidation with 2, 3-dichloro-5 and chemicalbook6-dicyanoquinone (DDQ). Adding a certain amount of weak acid in oxidative dehydrogenation can shorten the reaction time from 15h to 8h, and the yield can be increased from 47% to 62%. The reaction conditions were mild, the separation was simple and the total yield was 45%
