Tris(hydroxymethyl)aminomethane is a sodium-free amino buffer base, reacts with H2CO3 in body fluids to reduce H2CO3, while generating HCO32-, which can take up hydrogen ions and correct acidemia, its effect is strong, and can penetrate cell membranes, often used in acute metabolic and respiratory acidemia.  


Tris(hydroxymethyl)aminomethane is widely used in acute metabolic and respiratory acidemia, as an alkaline buffer, which has a good buffer effect on metabolic acidosis and enzyme activity reactions. Tris is commonly used as a biological buffer, often with pH values of 6.8, 7.4, 8.0, 8.8. Its structural formula and pH value vary greatly with temperature. In general, for every degree increase in temperature, the PH value decreases by 0.03. Tris is widely used in the preparation of buffers in biochemical and molecular biology experiments. For example, TAE and TBE buffers commonly used in biochemical experiments (for the dissolution of nucleic acids) require Tris. Because it contains amino groups, it can be condensed with aldehydes. The aqueous pH of Tris base is about 10.5, and a buffer of the pH value can be obtained by adding hydrochloric acid to adjust the pH value to the desired value. However, attention should be paid to the influence of temperature on the pKa of Tris. Since the Tris buffer is a weakly alkaline solution, DNA is deprotonated in such a solution, improving its solubility. EDTA is often added to Tris hydrochloric acid buffers to make "TE buffers", which are used for DNA stabilization and storage. If the pH adjusted acid solution is replaced by acetic acid, a "TAE buffer" (Tris/Acetate/EDTA) is obtained, and a "TBE buffer" (Tris/Borate/EDTA) is obtained. These two buffers are commonly used in nucleic acid electrophoresis experiments. TAE and TBE prepared from Tris are the most commonly used reagents for DNA electrophoresis, and TE (pH8.0) is mainly used to dissolve DNA (TE is a combination of Tris and EDTA). Tris buffers are not only widely used as solvents for nucleic acids and proteins, but also have many important uses. It is used for protein crystal growth under different pH conditions. The low ionic strength of Tris buffer can be used for the formation of intermediate fibers of lamin in C. legans, and Tris is also one of the main components of protein electrophoresis buffer. In addition, Tris is an intermediate in the preparation of surfactants, vulcanization accelerators and some drugs, and it is often used as a titration standard.
