General method for the preparation of intermediates 22-34

General procedure: Anhydrous piperazine (6 mmol) was dissolved in tetrahydrofuran (15 mL) under reflux; once piperazine was fully dissolved, benzyl chloride (1 mmol) was added drop-wise. The reaction mixture was then refluxed for 4h monitoring by TLC. The stirring mixture was cooled and then filtered. The obtained solid (excess of piperazine unreacted) was washed with THF (3 mL) and then with EtOAc (3 mL). The combined organic layers were concentrated in vacuo and then dissolved with basic water (NaOH 1 M) reaching pH>12. The aqueous layer was extracted with dichloromethane (25 mL) and then EtOAc (25 mL) keeping a pH>12. The organic layers were combined, dried over Na2SO4 and concentrated under vacuum. The crude product was purified by silica gel flash column chromatography on silica gel (DCM: MeOH 100:0 v/v, increasing to 90:10 v/v then DCM: MeOH: Et3N 90:9:1 v/v) to give the pure product.
