Reaction Conditions:

with bromine in diethyl ether at 0 - 20; for 17 h;

Steps:

3-(5-Bromo-2-thienyl)-3-[(2,2,2-trifluoroacetyl)amino]propanoicAcid (6)

To a suspension of 5 (8.4 g, 31.43 mmol, 1.0 equiv) in Et2O (250 mL)was added dropwise a solution of Br2 (2.08 mL, 40.87 mmol, 1.3equiv) at 0 °C. After stirring at rt for 17 h, the reaction mixture wasthen quenched with sat. aq Na2S2O3. The aqueous mixture was extractedseveral times with Et2O. The combined organic layers werewashed with brine, dried (MgSO4), filtered, and concentrated underreduced pressure to afford 6 as a light beige powder; yield: 10.85 g(quant.); mp 166-168 °C.IR (KBr): 3500-3300, 3290, 3091, 2422, 1703, 1550, 1433, 1218, 1161,1186, 800, 679 cm-1.1H NMR (DMSO-d6, 400 MHz): = 12.57 (s, 1 H), 10.08 (d, J = 8.0 Hz, 1H), 7.10 (d, J = 3.8 Hz, 1 H), 6.89 (d, J = 3.8 Hz, 1 H), 5.39 (q, J = 7.8 Hz,1 H), 2.98-2.91 (m, 2 H).13C NMR (DMSO-d6, 101 MHz): = 170.9, 155.6 (q, J = 36.6 Hz), 145.4,130.1, 125.9, 115.8 (q, J = 288.1 Hz), 110.4, 46.0, 39.0.19F NMR (DMSO-d6, 376 MHz): = -74.38.
