General procedure: Metformin hydrochloride 1 (1.0 mmol), sodium hydroxide (20 times) and tetrahydrofuran (THF, 10 mL) were added to the reactor, and cyanobenzene 2 (1.0 mmol) was slowly added to the above mixture at 80°C.Keep the reaction at the same temperature.After the reaction was monitored by TLC, the reaction solution was quenched with water (30 mL), extracted with ethyl acetate (4×10 mL), the organic layer was washed with saturated brine, dried over anhydrous sodium sulfate, filtered and concentrated to remove the solvent, and the resulting crude product was separated by column chromatography (Silica gel ~ 400 mesh, eluent: methanol/dichloromethane = 1/60 ~ 1/50), a total of 202 mg of compound 3 was obtained, with a yield of 79%, a white solid.
