Into a 250-mL round-bottom flask, was placed pyridin-2 -amine (4 g, 42.50 mmol, 1.00 equiv), AcOH (752 mg, 12.52 mmol, 0.52 equiv), butyl prop-2-enoate (3.7 g, 28.87 mmol, 1.20 equiv). The resulting solution was stirred overnight at 70 oC. Potassium hydroxide (3.37 g, 60.06 mmol, 2.50 equiv), water (10 mL) was added. The resulting solution was stirred overnight at 25°C. The resulting mixture was concentrated under vacuum. The resulting mixture was washed with DCM (200 mL x 6). The solids were filtered out. The filtrate was concentrated to give 4 g (57%) of 3 -(pyridin-2 -ylamino)propanoic acid as brown oil. MS (ES, m/z) [M+H]+: 167.
