Reaction Conditions:
Stage #1: 4-acetyl-1-(t-butoxycarbonyl)piperidinewith ammonium acetate;acetic acid in methanol at 25; for 2 h;
Stage #2: with sodium cyanoborohydride in methanol at 25; for 16 h;
Steps:
1.3 Synthesis of tert-butyl 4-(1-aminoethyl)piperidine-1-carboxylate
To a solution of tert-butyl 4-acetylpiperidine-1-carboxylate (7.9 g, 34.8 mmol) in MeOH (100 mL) was added NH4OAc (10.72 g, 139.2 mmol) and AcOH (1 mL). The mixture was stirred at 25 C for 2 h, then was added NaBH3CN (2.63 g, 41.76 mmol). The mixture was stirred at 25 C for 16 h. The mixture was evaporated and 2N NaOH (50 mL) was added to the residue. The solution was extracted with DCM (100 mL x 3 ). The DCM layer was washed with H2O and brine dried and evaporated to give the tert-butyl 4- (1-aminoethyl)piperidine-1-carboxylate (7.93 g, yield: 100 %)
