Steps:

1 Step 1: Synthesis of 6-chloro-4-methoxy-lH-pyrazolo [3, 4-d] pyrimidine:

To a solution of 4,6-dichloro-lH-pyrazolo[3,4-d]pyrimidine 1 (330 mg, 1.74 mmol) in methanol (10 mL) was added potassium carbonate (240 mg, 1.74 mmol). After the mixture was stirred at rt for 24 h, the reaction was quenched with saturated ammonium chloride (30 mL) solution and extracted with EA (20 mL x 3). The combined organic layer was dried over sodium sulfate (30 g), filtered and concentrated. The residue was purified by silica gel chromatography (eluent: EA/n-Hex = 1/10) to afford 120 mg of the title compound. LC-MS (Method A) (ESI+): m/z 185 (M+H)+; 1H-NMR (300 MHz, CDCb) d 10.69 (br s, 1H), 8.09 (s, 1H), 4.19 (s, 3H).
