Step 3; 1-N-tert-Butoxycarbonyl-4-(4-fluorobenzamido)piperidine: To a solution of 1-N-tert-butoxycarbonyl-4-aminopiperidine (430 mg, 2.4 mmol) in dichloromethane (30 mL) was added 4-fluorobenzyl chloride (0.26 mL) at 0° C. The reaction mixture was stirred at ambient temperature for 1 hour and subjected to standard extractive work up. The solvent was removed and the residue was purified by flash column chromatography on silica gel (petroleum ether-ethyl acetate 3:1) to give the title compound as a white solid (670 mg, 97% yield). 1H NMR (300 MHz, CDCl3) δ 7.77 (m, 2H), 7.14 (t, 2H), 5.90 (d, J=7.2 Hz, 1H), 4.10 (m, 3H), 2.91 (m, 2H), 2.03 (m, 2H), 1.39 (s, 9H), 1.35 (m, 2H); ESI-MS m/z=323.3 (M+H).
