Step 1: To a suspension of 2-chloro-3-nitrobenzaldehyde (CAS No. 58755-57-0, 9.4 g, 50.6 mmol) and K2CO3 (7.7 g, 55.7 mmol) in DMF (80 mL) was added dropwise methyl 2-mercaptoacetate (5.48 g, 51.6 mmol) at 0-5 °C. The resulting mixture was stirred at RT for 12 h. The reaction mixture was diluted with water. The precipitates formed were filtered, washed with water, and dried to afford methyl 7-nitrobenzo[b]thiophene-2-carboxylate as a yellow solid (11.5 g, 95%). LC/MS (ES+): m/z calculated for C10H7NO4S: 237.0; found: 238.4 [M+H] NMR (400 MHz, CDCb): 6 8.53 (d, = 8.0 Hz, 1H), 8.23 (d, = 8.0 Hz, 1H), 8.19 (s, 1H), 7.61 (t, = 8.0 Hz, 1H), 3.99 (s, 3 H).
