以2,2'-(5-甲基-1,3-亚苯基)二乙腈和碘甲烷为原料合成5,α,α,α',α'-五甲基-1,3-苯二乙腈的一般步骤:将3,5-双(氰甲基)甲苯(800g,4.70mol)、甲基碘(2935.2g,20.68mol)和二甲基甲酰胺(11.20L)的混合物冷却至0℃至5℃。在1至1.5小时内分批加入60%氢化钠在矿物油中的分散体(864.2g,36.0mol)。随后,将反应混合物逐渐升温至室温并持续搅拌2至2.5小时。反应进程通过薄层色谱(TLC,展开剂为正己烷:乙酸乙酯=7.5:2.5)进行监测。确认反应完成后,加入乙酸乙酯以淬灭过量的氢化钠。将反应混合物用水稀释,并用二氯甲烷(3×5L)进行萃取。合并有机相,用盐水(5L)洗涤。有机相在室温下用活性炭脱色1小时,随后在40℃至45℃下减压浓缩至干。所得残余物于70℃至75℃下溶解于四氯化碳(2400mL)中,并逐渐冷却至10℃至15℃,搅拌1小时。进一步将溶液冷却至-5℃至0℃,继续搅拌3小时。最后,过滤搅拌的悬浮液,用预冷的四氯化碳(500mL)洗涤滤饼,并于40℃至45℃下真空干燥,得到3,5-双(1-氰基-1-甲基乙基)甲苯(748.0g,产率70.3%)。
