将市售的外消旋1,1’-联-2-萘酚与市售的(S)-脯氨酸以摩尔比为1:1的比例在乙腈中混合(乙腈的用量为外消旋的1,1’-联-2-萘酚重量的15倍),加热回流3小时,冷至室温,有白色固体析出,抽滤得到白色固体和乙腈滤液。白色固体用乙醇加热溶解,自然冷却至室温,静置,析出块状无色透明晶体(分子包合物结晶,Mp220–225℃,1HNMR(DMSO-d6):δ9.32(s,4H,disappearedafteradding D2O),7.85(d,J=8.1Hz,8H),7.33(d,J=9Hz,4H),7.25–7.14(m,8H),6.93(d,J=8.1Hz,4H),3.66(t,J=8.1Hz,1H),3.24-3.16(m,1H),3.05-2.96(m,1H),2.02-1.91(m,2H),1.80-1.66(m,2H).13CNMR(DMSO-d6):δ171.6,152.1,133.3,128.2,127.6,126.9,125.0,123.8,121.6,117.7,113.9,59.9,44.7,28.7,23.4.),抽滤,室温下,将该分子包合物结晶加入到乙酸乙酯-水两相体系(有机相和水相体积比2:1)中搅拌1小时,分液,水相用乙醚萃取,合并有机相,有机相经干燥、蒸发和残余物用甲苯重结晶得到对映纯(S)-1,1’-联-2-萘酚块状晶体,总收率:68%,100%ee,m.p.208-210℃;[α]D20=-36.5(c1,inTHF).
水相蒸发浓缩至干后,用乙醇重结晶,回收(S)-脯氨酸,收率:92%。
乙腈滤液经蒸发浓缩至干、剩余物乙醚提取、乙醚提取液蒸发、蒸发后的残余物在甲苯中重结晶两次,得到对映纯R-1,1’-联-2-萘酚,总收率:41%,100%ee,m.p.208-210℃;[α]D20=+36.4(c1,inTHF).
