以2,5-二溴吡啶和N,N-二甲基甲酰胺(DMF)为原料,按照通用方法C合成5-溴-2-吡啶甲醛。具体操作如下:将2,5-二溴吡啶(237 mg, 1 mmol)与DMF(115 μL, 1.5 mmol)混合反应。反应完成后,通过快速柱色谱法(洗脱剂:己烷/乙酸乙酯=10:1)纯化粗产物,得到目标化合物5-溴-2-吡啶甲醛(27b),为褐色固体(149 mg, 产率80%)。薄层色谱(TLC)分析显示Rf值为0.45(展开剂:己烷/乙酸乙酯=8:1)。核磁共振氢谱(1H NMR, 400 MHz, CDCl3)数据如下:δ 10.03 (1H, s), 8.85 (1H, dd, J = 0.8, 2.0 Hz), 8.02 (1H, ddd, J = 0.8, 2.0, 8.0 Hz), 7.85 (1H, dd, J = 0.8, 8.0 Hz)。核磁共振碳谱(13C NMR, 100 MHz, CDCl3)数据如下:δ 192.4, 151.7, 151.3, 140.0, 126.3, 122.8。所得光谱数据与文献报道一致。
