将2.2 mL乙酸酐(22 mmol)缓慢滴加到预先在高真空下用P4O10干燥2天的鸟苷(2 g,7.0 mmol)悬浮液中,该悬浮液含有三乙胺(7.7 mL,55.2 mmol)和N,N-二甲基氨基吡啶(92 mg,0.75 mmol)于27 mL乙腈中,反应在0℃下进行。搅拌混合物直至均匀,随后在室温下继续反应3小时。反应完成后,用甲醇(2.3 mL)淬灭反应。使用旋转蒸发器将反应液体积浓缩至原体积的1/3,随后缓慢加入乙醚以诱导细白色粉末沉淀。通过过滤收集产物,用乙醚洗涤,然后在50℃下用丙酮(30 mL)搅拌2小时。最终得到2.8 g(产率98%)的细白色粉末产物。产物经1H NMR(200 MHz, DMSO)和13C NMR(200 MHz, CDCl3)分析确认,质谱分析结果与理论值相符(C18H21N5O5 [M+H]+计算值:410.1306,实测值:410.1305)。
