以2-氟-4-溴甲苯为原料合成2-氟-4-溴溴苄的一般步骤:将N-溴代琥珀酰亚胺(NBS,1.07g,6.05mmol)和偶氮二异丁腈(AIBN,83mg,0.50mmol)溶解于四氯化碳(CCl4,10mL)中,随后加入2-氟-4-溴甲苯(1g,5.04mmol)。将反应混合物在100℃下加热回流13小时。反应完成后,将混合物冷却至室温,过滤收集生成的固体。用四氯化碳和水对滤液进行多次萃取。合并有机层,用无水硫酸钠干燥以除去残留水分。随后,在减压条件下浓缩有机层,得到4-溴-1-(溴甲基)-2-氟苯(1.2g,4.47mmol),产率为89%。产物经1H-NMR(300MHz,CDCl3)表征:δ7.51-7.41(m,1H),7.09(d,J=9.0Hz,1H),6.99(d,J=8.2Hz,1H),4.34(s,2H)。
