Steps:

20.1 Step 1: Preparation of Y-1

Add 4-nitroindole (4000mg), NH(CH3)2(13.58ml, 2mol/lin MeOH), HCHO (2.2ml, 37%), ZnCl2(5048mg) and 10ml Methanol. After stirring at room temperature for 2 h, additional NH(CH3)2 (2.46ml), HCHO (0.5ml) were added. After stirring for 4 hours, LC-MS showed that the raw materials were consumed completely. NaOH solution was added to adjust the pH to about 13, EA was added to dilute, and the insoluble solid was removed by filtration. The filtrate was extracted with EA, and after the sodium sulfate solid was dried, the solvent was spin-dried to obtain a yellow solid, which was then beaten with PE/EA 10:1 to obtain 4250 mg of a yellow solid.
