Weigh 2-(10-hydroxydecyl)-3,4,5-trimethoxytoluene (3.38g, 0.01mol) and dissolve in 5mL of acetone. Add 0.05 mL of concentrated nitric acid, slowly drop 10 mL of an aqueous solution containing 0.015 mol of ammonium persulfate through a constant pressure dropping funnel, and complete the drop within 10 minutes, and react at 50 °C. TLC monitor every half hour, react for 2 hours, stop the reaction, add 10 mL of water to quench the reaction, and extract with 30 mL of dichloromethane three times. The organic layer was washed 3 times with 15 mL of saturated sodium bicarbonate aqueous solution, and washed with pure water until the pH of the aqueous phase was about 6. Combine the organic phases, dry the organic phases with anhydrous sodium sulfate. Finally, it is filtered, spin-dried, and recrystallized with n-hexane to obtain red needle-like crystals, that is, the total yield of idebenone is 98%.
