A solution of 52.300 mmol of C-(3-benzyloxypyridin-2-yl)methylamine [88423-14-7] (Example 2A) in 35 ml of formic acid is heated to reflux for 3 hours. The reaction mixture is cooled and adjusted cautiously to pH 8 with approx. 100 ml of ammonium hydroxide solution (25% in water) with ice bath cooling. The mixture is diluted with 100 ml of water and the aqueous phase is extracted repeatedly with dichloromethane (3 x 250 ml). The combined organic phases are dried over sodium sulphate and concentrated by evaporation. 51 .590 mmol (99% crude yield) of the title compound are obtained from the residue as a brown oil. The material is used in the next step without further purification. Rf = 0.16 (ethyl acetate), Rt = 4.58.
