2-Bromo-6-fluoropyridine was synthesized from 2-bromopyridine via mixed acid nitration, nitro reduction, diazotization, and the introduction of fluorine atoms through the Schiemann reaction. This route simplifies the experimental procedure and avoids the problems of low yield and poor purity reported in the literature regarding the synthesis of the starting material 2-chloro-5-aminopyridine, the separation of intermediate products, and the purification of the final product. The optimal process conditions (reaction temperature, reaction time, and molar yield) for each step were as follows: nitration: 45℃, 2h, 41%; nitro reduction: reflux, 1h, 90%; diazotization: -5℃ to 0℃, 2h, 81.4%; Schiemann reaction: 130℃, 0.5h, with an overall yield of 51.6% for both steps. The synthetic reaction formula is shown in the figure below: Figure 2-bromo-6-fluoropyridine |
