在氩气保护下,将NaH(179 mg,4.10 mmol,55%油溶液)悬浮于THF(2 mL)中,冷却至0℃。依次加入(S)-环氧乙烷-2-基甲醇(S1, 200 mg,2.70 mmol)的THF溶液(4 mL)和苄基溴(0.340 mL,2.86 mmol)。将反应混合物加热回流搅拌19小时。反应完成后,用0.1 M磷酸盐缓冲液(pH 6.5)淬灭反应。用乙酸乙酯(3×)萃取产物,合并有机层,用饱和食盐水洗涤,无水硫酸钠干燥。减压浓缩后,残余物通过硅胶快速柱色谱纯化(洗脱剂:己烷/乙酸乙酯 = 10/1),得到(R)-苄氧甲基环氧乙烷(S21)为无色油状物(196 mg,44%)。1H NMR (300 MHz, CDCl3) δ = 2.62 (dd, J1 = 2.5 Hz, J2 = 5.0 Hz, 1H), 2.80 (dd, J1 = 3.0 Hz, J2 = 5.0 Hz, 1H), 3.19 (dddd, J1 = 2.5 Hz, J2 = J3 = 3.0 Hz, J4 = 5.5 Hz, 1H), 3.44 (dd, J1 = 5.5 Hz, J2 = 11.5 Hz, 1H), 3.77 (dd, J1 = 3.0 Hz, J2 = 11.5 Hz, 1H), 4.57 (d, J = 12.0 Hz, 1H), 4.61 (d, J = 12.0 Hz, 1H), 7.25-7.38 (m, 5H); 13C NMR (75 MHz, CDCl3) δ = 44.3, 50.8, 70.8, 73.3, 127.7, 128.4, 137.8; HRMS (EI) m/z 164.0825 (calculated for C10H12O2, M+ 164.0837).
