将2,5-二羟基苯甲酸(5.00g,32.5mmol)溶于甲醇(33mL)中,缓慢滴加浓硫酸(5mL)。反应混合物在回流条件下加热16小时。反应完成后,将混合物冷却至室温,并在真空条件下除去溶剂。残余物用饱和NaHCO3水溶液中和至pH 7。随后,用乙酸乙酯(50mL×3)萃取产物,合并有机层,用盐水(20mL)洗涤,无水MgSO4干燥,过滤后蒸发滤液,得到浅棕色粉末状产物2,5-二羟基苯甲酸甲酯(5.13g,收率94%)。产物结构经1H NMR(CDCl3)δ 10.35(s,1H),7.29(d,J = 3.2Hz,1H),7.02(dd,J = 8.7,3.2Hz,1H),6.89(d,J = 8.7Hz,1H),4.69(s,1H),3.95(s,1H);13C NMR(CDCl3)δ 170.1,155.8,147.6,124.0,118.5,114.7,112.1,52.4;HRMS(ESI):[M-H]-计算值为C8H7O4 167.0344,实测值167.0359确认。
