方案1,步骤B:制备(R)-(-)-5-(羟甲基)-2-吡咯烷酮(3) 向含有(R)-5-氧代吡咯烷-2-羧酸甲酯(中间体2,13.2 g)的甲醇(100 mL)溶液中,在0℃下分批加入硼氢化钠(10.5 g,278 mmol)。反应混合物在0℃下持续搅拌直至反应完成,随后加入乙酸(3 mL)以淬灭反应。反应混合物经浓缩后,残余物通过硅胶柱层析纯化,采用甲醇-氯仿(1:9,v/v)作为洗脱剂,得到目标产物(12.9 g,收率97%)为无色固体。薄层色谱(TLC)显示Rf值为0.33(展开剂系统:甲醇-氯仿,1:9,v/v);核磁共振氢谱(1H-NMR,CDCl3)δ:7.17(s,1H),3.92(s,1H),3.85-3.75(m,1H),3.64-3.40(m,2H),2.42-2.35(m,2H),2.2-2.05(m,1H),1.88-1.7(m,1H)。
