将吲哚-3-基乙酸(1.00 g,5.71 mmol)溶于25 mL乙醇中,加入浓硫酸(1.0 mL,18.4 mmol)。将反应混合物在回流条件下搅拌2小时。反应完成后,将混合物在冰浴中冷却至0℃,随后用2 N氢氧化钠溶液中和。减压蒸发除去有机溶剂,将残余物在二氯甲烷和水之间分配。水相用二氯甲烷萃取两次,合并有机相,用饱和食盐水洗涤,无水硫酸钠干燥,过滤后减压浓缩,得到吲哚-3-醋酸乙酯1.02 g(5.02 mmol,收率88%),为棕色油状物。产物经核磁共振氢谱(1H NMR,300 MHz,CDCl3)确认:δ 8.09(br.s, 1H), 7.63(d, J = 7.69 Hz, 1H), 7.35(d, J = 7.69 Hz, 1H), 7.04-7.25(m, 3H), 4.17(q, J = 7.14 Hz, 2H), 3.77(s, 2H), 1.26(t, J = 7.14 Hz, 3H)。超高效液相色谱-质谱联用(UPLC/MS)分析显示保留时间为1.58分钟,低分辨质谱(LRMS)测得m/z 204([M+1]+)。
