A mixture of tert-butyl piperazine-1-carboxylate (0.931 g,5 mmol),bromoacetonitrile (0.348 mL,5.00 mmol) and potassium carbonate (1.037 g, 7.50 mmol) in DMF (20 mL) was stirred at rt for 18 hr. The reaction mixture was partitioned between EtOAc (75 ml) and water (75 ml). The organic layer was washed with 10% LiCl solution (2 x 75 ml) and brine (75 ml). After drying (Na2S04) and filtration, the organic layer was concentrated to afford tert-butyl 4-(cyanomethyl)piperazine-1-carboxylate (1.08 g,4.79 mmol, 96 % yield) as a dark yellow solid. NMR (400MHz, chloroform-d) d 3.53 (s, 2H), 3.51- 3.45 (m,4H), 2.60 -2.47 (m,4H), 1.47 (s, 9H).
