2.2. Preparation of 2-alkyl/aralkyl/heterocyclyl sulfanyl-5-amino/methyl-1,3,4-thiadiazoles (1-22)
General procedure: The title compounds were prepared by following the Scheme 1. To a cold solution (10° C) of potassium hydroxide (10 mmol) in absolute ethyl alcohol (50 mL), 5-methyl/amino-1,3,4-thidiazole-2-thiol (5 mmol) was added in small lots. The reaction mixture was stirred and cooled to 10° C followed by the addition of a solution of alkyl iodide/bromide or allyl bromide/benzyl chloride (5.5 mmol) or dibromopropane (2.5 mmol) in ethyl alcohol (20 mL). The reaction mixture was stirred at ambient temperature for 7-8 h and thin layer chromatography was used to monitor the progress of reaction by using the solvent system of 1:1 mixture of hexane and ethyl acetate. The solid product was obtained which was filtered followed bywashing with water and recrystallization from ethanol/water.
