EPN is commercially produced via a process that begins with the preparation of O-ethyl phenylphosphonochloridothioate by reacting phenylphosphonothioic dichloride with ethanol in the presence of triethylamine in toluene. The key nucleophilic substitution follows, coupling the chloridothioate with 4-nitrophenol under basic conditions to form the aryl ester linkage. Final purification via vacuum distillation or crystallisation from isopropanol yields EPN.
