Reaction Conditions:

Stage #1:carbon disulfide with ammonia;triethylamine in ethanol;water at 20;
Stage #2: with iodine in ethanol;water;acetonitrile for 0.5 h;

Steps:

4.2. Synthesis of thiosemicarbazones (1a-1k, 2a-2h and 3a-3g)[41-43]
General procedure: Primary amine (5mmol) was added to 15mL of the mixed solution of ethanol and water (1:1). Then, CS2 (5eq) and triethylamine (1eq) were added to the solution, and the solution was stirred at room temperature overnight. Iodine elemental (dissolved in acetonitrile, 1eq) was slowly added to the mixture and stirred until yellow precipitated out. After further reaction for 30min, the organic solvent was concentrated and extracted with n-hexane. The organic phase was washed with saturated brine and dried over anhydrous sodium sulfate. Concentrating on the organic phase, and 10mL of isopropanol was added. In an ice bath, hydrazine hydrate was slowly added to isothiocyanate-containing isopropanol and stirred for 10min until the precipitate was completely precipitated. The precipitate was filtered and dried under vacuum.
