4-羟基-3-甲氧基苯甲酸乙酯的合成:在配备氮气入口和磁力搅拌子的圆底烧瓶中,加入香草酸(10 g,59.49 mmol)的无水乙醇(400 mL)溶液。向上述溶液中缓慢加入浓盐酸(600 mg,6.11 mmol),随后滴加浓硫酸。将反应混合物在回流条件下搅拌48小时。反应完成后,使用旋转蒸发仪除去溶剂。向残余物中加入去离子水(100 mL),通过分液漏斗分离并弃去绿色油状物。将所得产物在真空下干燥,得到4-羟基-3-甲氧基苯甲酸乙酯(11.45 g,收率98%)。产物经1H NMR(400 MHz,CDCl3)表征:δ 7.62(dd,J = 8.5, 2.1 Hz,1H),7.53(d,J = 1.8 Hz,1H),6.91(d,J = 8.6 Hz,1H),4.33(q,J = 7.1 Hz,2H),3.91(s,3H),1.36(t,J = 7.3 Hz,3H)。HPLC-MS分析显示:预期分子离子峰m/z 197(MH+),实测m/z 197。
