A four carbon dicarboxylic acid that is an intermediate in the citric acid cycle and glucogenesis. It has been shown to inhibit succinate dehydrogenase.

This tricarboxylic acid cycle intermediate (FWfree-acid = 132.07 g/mol; CAS 328-42-7; Abbreviation: OAA), also known as 2-oxosuccinic acid, 2- oxobutanedioic acid, and oxalacetic acid, is the a-keto acid transamination product of aspartate. Solution Chemistry: OAA has pKa values of 2.22 and 3.67, and the OH group in enol form has a pKa value of 13.03. Note that it is relatively unstable in aqueous solutions and undergoes b-decarboxylation. The half-life of oxaloacetate at pH 7 and 25°C is between 30 and 60 minutes. Equilibrium exchange studies of malate dehydrogenase were made at 1°C to minimize the decarboxylation rate of oxaloacetate. While the solid dicarboxylic acid exists primarily as the enol, aqueous oxaloacetate, at pH 6 to 10, is roughly 82-88% keto, 7-10% enol, and 5-8% gem-diol. At pH 1.3, most of the oxaloacetic acid is present as the gem-diol (~81% vs. 13% keto and 6% enol). In diethyl ether, more than 90% is found as the enol. The rate of decarboxylation is maximum at pH 3–4; this rate is accelerated in the presence of metal ions, with the pH optimum shifting to 5–6.5. Metabolism: Oxaloacetate is a product of the reactions catalyzed by the malate dehydrogenases, malate oxidase, D- and L-aspartate oxidases, methylmalonyl-CoA carboxyltransferase, aspartate aminotransferase, aspartate:phenylalanine aminotransferase, 4-methyloxaloacetate esterase, citrate lyase, oxalomalate lyase, citryl-CoA lyase, L (+) - and D (–) -tartrate dehydratases, threo- and erythro-3-hydroxyaspartate ammonia-lyases, and pyruvate carboxylase. It is a substrate for lactate:malate transhydrogenase, citrate (si) - and (re) -synthases, decylcitrate synthase, 2-methylcitrate synthase, ATP citrate synthase pyridoxamine:oxaloacetate aminotransferase, glycine:oxaloacetate aminotransferase, oxaloacetase, oxaloacetate decarboxylase, phosphoenol-pyruvate carboxylase, phosphoenolpyruvate carboxy-kinases, and oxaloacetate tautomerase. It is also an alternative substrate/product for many other aminotransferases, homocitrate synthase, and opine dehydrogenase.
