5-Amino-2-methoxyphenol reacts with ninhydrin to yield tetrahydroindeno[1,2-b]indolones. | [Synthesis]
Commercially available 2-methoxy-5-nitrophenol (40 g, 236.5 mmol) was dissolved in methanol (300 mL) under argon protection and palladium/carbon catalyst (3 g) was added. The suspension was vacuum degassed and purged repeatedly with hydrogen several times. Subsequently, the reaction mixture was stirred at room temperature under hydrogen pressure (40 psi) for 24 hours. After completion of the reaction, the catalyst was removed by filtration and the filtrate was concentrated to afford the intermediate 2-methoxy-5-aminophenol (25 g, 76% yield) as a yellow solid. Electrospray ionization mass spectrometry (ESI-MS) showed [M + 1] peak of 140.1, which was consistent with the calculated value. Elemental analysis (C7H9NO2) calculated value: 139.06. | [References]
