순도시험

  (1) 굴절률 : 이 품목의 굴절률 은 1.525～1.529이어야 한다.

  (2) 용상 : 이 품목 1.0g을 이소옥탄 10mL에 녹일 때, 그 액은 황색이며 맑아야 한다 .

  (3) 흡광비 : 이 품목의 이소옥탄용액(1→100,000)의 파장 248.5nm, 253.5nm 및 269.5nm에 있어서의 흡광도 A₁, A₂ 및 A₃를 측정할 때, A₂/A₁은 0.69～0.73, A₂/A₃는 0.74～0.78이어야 한다. 또한 이 품목의 이소옥탄용액(1→10,000)의 파장 284.5nm 및 326.0nm에서의 흡광도 A₄ 및 A₅를 측정할 때, A₄/A₅는 0.28～0.34이어야 한다.

  (4) 납 : 이 품목 5.0g을 취하여 원자흡광광도법 또는 유도결합플라즈마발광광도법에 따라 시험할 때, 그 양은 2.0ppm 이하이어야 한다.

  (5) 메나디온 : 이 품목 20mg에 에탄올․물의 혼액(1 : 1) 0.5mL를 가하여 섞은 다음 1-페닐-3-메칠-5-피라졸론의 알콜용액(1→20) 1방울 및 암모니아수 1방울을 가한 다음 2시간 방치할 때, 청자색을 나타내어서는 아니 된다.

확인시험

  (1) 이 품목의 이소옥탄용액(1→100,000)은 흡광도측정법에 따라 시험할 때, 따로 비타민K₁ 표준품을 같은 방법으로 측정하였을 때와 동일한 자외부흡수스펙트럼을 나타내어야 한다.

  (2) 이 품목 50mg을 에탄올 10mL에 녹인 다음 알콜성10%수산화칼륨시액 1mL를 가할 때, 액의 색은 청색을 나타내며 방치하면 자색을 거쳐 갈색으로 변한다.

  (3) 이 품목 50mg을 메탄올․에탄올의 혼액(1 : 1) 10mL에 녹이고, 쓸 때 히드로설파이트나트륨 0.75g을 따뜻한 물 2mL에 녹인 액을 가하여 세게 흔들어 섞을 때 액의 황색이 없어진다.

정량법

  이 품목 약 0.1g을 정밀히 달아 100mL 메스플라스크에 넣고 이소옥탄을 가하여 녹인 다음 이소옥탄을 가하여 100mL로 한 후 이 액 10mL를 취하여 이소옥탄으로 100mL로 희석하고 다시 이 액 10mL를 취하여 이소옥탄으로 100mL로 한 것을 시험용액으로 한다. 따로, 비타민K₁ 표준품 약 0.1g을 정밀히 달아 시험용액과 동일조작 한 것을 표준용액으로 한다. 이소옥탄을 대조액으로 하여 액층 1cm, 파장 248.5nm부근의 극대흡수파장에서 시험용액 및 표준용액의 흡광도를 측정하여 다음 계산식에 따라 함량을 구한다.

  차광한 밀봉용기에 넣고 보존하여야 한다.

개요

Vitamin K₁ is a fat-soluble, dietary nutrient that is essential for the synthesis of proteins important for blood-clotting, bone metabolism, and cell growth. It is found in the photosynthetic tissues of green, leafy plants, where it acts as an electron acceptor forming part of the electron transport chain of Photosystem I. Vitamin K₁ also serves as a precursor to vitamin K₂ and is reported to exhibit anticancer activity in various cell lines.

일반 설명

Vitamin K₁, also known as phylloquinone, consists of naphthalenoid and phytyl groups. Phylloquinone is present abundantly in photosynthetic plants and is a major dietary source. It is a fat-soluble vitamin.

Phylloquinone is a dietary component essential for normal biosynthesis of several factors required for clotting of blood; as a therapeutic drug used to correct bleeding tendency; and as a food supplement.

Purification Methods

Vitamin K1 is a yellow viscous oil, which can be distilled at high vacuum practically unchanged. It is insoluble in H2O, but soluble in common organic solvents. Store it in the dark under N2 as it is oxygen sensitive. It has  1cm 328 at 248nm. [Fieser et al. J Am Chem Soc 61 2557 1939, Hirschmann et al. 비 호환성

Phylloquinone is photosensitive; decomposes in sunlight and is destroyed by alkali hydroxides and reducing agents such as hydrideds and active metals. Incompatible with oxidizers (chlorates, nitrates, peroxides, permanganates, perchlorates, chlorine, bromine, fluorine, etc.); contact may cause fires or explosions. Keep away from alkaline materials, strong bases, strong acids, oxoacids, epoxides.

폐기물 처리

It is inappropriate and possibly dangerous to the environment to dispose of expired or waste drugs and pharmaceuticals by flushing them down the toilet or discarding them to the trash. Household quantities of expired or waste pharmaceuticals may be mixed with wet cat litter or coffee grounds, double-bagged in plastic, discard in trash. Larger quantities shall carefully take into consideration applicable DEA, EPA, and FDA regulations. If possible return the pharmaceutical to the manufacturer for proper disposal being careful to properly label and securely package the material. Alternatively, the waste pharmaceutical shall be labeled, securely packaged and transported by a state licensed medical waste contractor to dispose by burial in a licensed hazardous or toxic waste landfill or incinerator.
